How to Read a SS-31 (Elamipretide) Certificate of Analysis

A COA is only meaningful if the identifiers on it match the compound you ordered. Four checks, in order.

  1. 1. Identity โ€” does the formula match? The COA header should state C32H49N9O5 and a molecular weight near 639.8 g/mol, with CAS 736992-21-5. A mismatch means you are not looking at a COA for this compound.
  2. 2. Purity โ€” HPLC chromatogram, not just a numberA printed “> 99.0%” with no chromatogram is a marketing claim. Demand the actual HPLC trace: a single dominant peak, labelled retention time, and an integration table. Shoulders or unlabelled side peaks are unreported impurities.
  3. 3. Mass confirmation โ€” does MS agree with the formula?Mass spectrometry should report an observed mass consistent with the expected 639.8 g/mol. Peptides are frequently reported as multiply charged ions, so the raw m/z value will be lower than the neutral mass. An unexplained offset is a red flag.
  4. 4. Provenance โ€” who signed it, and for which batch?A valid COA names the testing laboratory, carries a batch or lot number that matches your vial label, and is dated. A vendor-produced PDF with no lab identity and no batch number is not third-party evidence.

Mechanism of Action

SS-31 (H-D-Arg-Dmt-Lys-Phe-NHโ‚‚, where Dmt is 2,6-dimethyl-L-tyrosine) is a synthetic aromatic-cationic tetrapeptide from the Szeto-Schiller series that selectively accumulates in the inner mitochondrial membrane. Its targeting is driven by electrostatic attraction between its cationic residues (D-Arg, Lys) and the negatively charged cardiolipin phospholipid, combined with hydrophobic insertion into the lipid bilayer. Cardiolipin is almost exclusively found in the inner mitochondrial membrane and is essential for cristae architecture, electron transport chain (ETC) supercomplex assembly, and cytochrome c function.

The primary mechanism is cardiolipin protection and stabilization. Under oxidative stress, cytochrome c dissociates from cardiolipin and converts into a peroxidase that peroxidizes cardiolipin, disrupting ETC supercomplexes, collapsing cristae structure, and unleashing a vicious cycle of electron leak and ROS production. SS-31 binds cardiolipin and prevents this cytochrome c conformational shift, preserving its electron-carrier function while inhibiting its pathological peroxidase activity. This is fundamentally different from direct antioxidant mechanisms โ€” SS-31 reduces mitochondrial ROS indirectly by improving electron transport efficiency and reducing electron leak, rather than by scavenging free radicals.

Downstream effects include: preservation of cristae morphology after ischemic or metabolic stress; enhanced assembly and stability of ETC complexes Iโ€“IV and their supercomplexes; restored oxygen consumption and ATP synthesis efficiency; and modulation of cardiolipin-dependent membrane surface electrostatics that tunes how proteins associate with the membrane. The use of D-amino acids (D-Arg, Dmt) confers resistance to proteolytic degradation.

Research History & Context

SS-31 (elamipretide, formerly MTP-131/Bendavia) was developed by Drs. Hazel Szeto and Peter Schiller at Cornell University in the early 2000s as part of a systematic search for mitochondria-targeted peptides. Stealth BioTherapeutics advanced the compound through an extensive clinical program spanning heart failure (PROGRESS-HF), primary mitochondrial myopathy (MMPOWER-3), Barth syndrome (TAZPOWER), and dry age-related macular degeneration (ReCLAIM-2).

The clinical history has been mixed: PROGRESS-HF, MMPOWER-3, the randomized portion of TAZPOWER, and ReCLAIM-2 all failed to meet their primary endpoints. However, on September 19, 2025, the FDA granted accelerated approval to elamipretide hydrochloride (Forzinity) for improving muscle strength in Barth syndrome patients โ€” the first approved therapy for this rare mitochondrial disorder and the first FDA-approved cardiolipin-directed agent. Continued approval may be contingent on confirmatory trial results. A 2021 mechanistic study in older adults demonstrated that a single IV infusion improved in vivo skeletal muscle mitochondrial ATP production, reversing age-related declines โ€” one of the strongest human bioenergetic signals for any mitochondrial-targeted compound.

Storage & Handling

Store lyophilized SS-31 at -20ยฐC in a light-protected, desiccated container. The peptide's D-amino acid residues (D-Arg, Dmt) confer significant resistance to standard peptidases. Stability data is well-characterized from the pharmaceutical development program; lyophilized material is stable >24 months at -20ยฐC.

After reconstitution with bacteriostatic water, SS-31 should be refrigerated at 2โ€“8ยฐC and used within 21 days. The peptide is water-soluble and forms a clear, colorless solution. Do not freeze reconstituted solutions. For research protocols using larger doses (20โ€“40 mg), reconstitute at higher concentrations (e.g., 1.0 ml BAC water for 10mg = 10 mg/ml) to manage injection volume. SS-31 has been researched both as monotherapy and in combination with MOTS-c for complementary mitochondrial effects โ€” SS-31 targeting membrane structure/cardiolipin and MOTS-c targeting nuclear-mitochondrial crosstalk via AMPK/SIRT1.

Reconstitution & Research Dosing

For SS-31 (Elamipretide), the standard laboratory reconstitution is 2.0 ml BAC water for 10mg vial. A typical research study window uses 4โ€“40 mg / daily or 2โ€“3ร— weekly (wide research range).

For a 10mg SS-31 research vial, add 2.0 ml BAC water (5.0 mg/ml). Research protocols vary widely โ€” from 4 mg daily (targeted mitochondrial support) to 40 mg 2โ€“3ร— weekly (more intensive protocols). Due to the wide dose range, researchers should select reconstitution volumes appropriate to their protocol: for 4 mg doses, 2.0 ml BAC water yields manageable injection volumes; for 40 mg doses, reconstitute with 1.0 ml BAC water (10 mg/ml) and use a 3ml syringe. SS-31 is a cationic peptide; some protocols use preservative-free sterile water for injection when same-day administration of the entire vial is planned.

SS-31 FAQ

QWhat is the molecular formula of SS-31 (Elamipretide)?

SS-31 (Elamipretide) has the molecular formula C32H49N9O5 and a molecular weight of approximately 639.8 g/mol. Its CAS registry number is 736992-21-5. It is a 4-residue peptide with the sequence D-Arg-2,6-dimethyl-Tyr-Lys-Phe-NH2.

QHow do you verify a SS-31 (Elamipretide) Certificate of Analysis?

Check four things in order. First, identity: the COA must state C32H49N9O5 and CAS 736992-21-5. Second, purity: require the actual HPLC chromatogram with retention time and integration table, not just a printed percentage. Third, mass confirmation: the mass spectrometry result must agree with the expected molecular weight of 639.8 g/mol, allowing for multiply charged ions. Fourth, provenance: the COA must name the testing laboratory and carry a batch number matching your vial label.

QWhat purity should SS-31 (Elamipretide) be?

Research-grade SS-31 (Elamipretide) should meet a purity target of > 99.0%, confirmed by third-party HPLC analysis rather than a vendor self-declaration. A purity claim without an accompanying chromatogram carries no evidential weight.

QWhat is the half-life of SS-31?

The reported half-life of SS-31 is ~2 hours (plasma). Half-life figures come from published research models and vary with species, route and formulation, so treat them as a reference range rather than a fixed value.
