Mechanism of Action
Epithalon (Ala-Glu-Asp-Gly) is a synthetic tetrapeptide designed to mimic the active region of epithalamine, a polypeptide extract from the pineal gland. Its primary proposed mechanism is activation of telomerase — the enzyme responsible for extending telomeres, the protective caps at chromosome ends that shorten with each cell division and serve as a molecular clock of cellular senesence.
In addition to telomerase activation, epithalon has been shown to regulate melatonin and circadian rhythm gene expression, modulate pineal peptide secretion, influence the hypothalamic-pituititary axis, and reduce oxidative stress markers in aged animal models. Research from the St. Petersburg Institute of Bioregulation and Gerontology has reported lifespan extension and reduced age-related pathology in rodent and primate studies.
The peptide's effects appear to be cycle-dependent rather than continuous — a 10–20 day administration cycle repeated every 4–6 months is the typical research protocol, rather than ongoing daily administration.
Research History & Context
Epithalon was developed in the 1990s by Prof. Vladamir Khavinson at the St. Petersburg Institute of Bioregulation and Gerontology, as part of a broader program of peptide bioregulators derived from tissues of young animals. It has been studied in over 50 preclinical and clinical publications, primarily from Russian research institutions.
A 15-year observational study in elderly human subjects (published 2003–2018) reported reduced all-cause mortality and lower rates of age-associated disease in the epithalon-treated cohort compared to controls. However, independent replication outside of the originating research group remains limited, and rigorous double-blind trials have not been completed by Western research institutions as of 2026.
Storage & Handling
Store lyophilized epithalon at -20°C in a dark, dry environment. Stability data is more limited than for other research peptides; conservative protocols recommend use within 12 months of lyophilization even when frozen.
After reconstitution with bacteriostatic water, epithalon should be refrigerated at 2–8°C and used within 14 days — a shorter window than many other peptides due to limited stability data. Some researchers reconstitute with sterile water for injection (rather than BAC water) for immediate use only, discarding any remaining solution. Freeze-thaw of reconstituted epithalon is not recommended.
Reconstitution & Research Dosing
For Epithalon (Epitalon), the standard laboratory reconstitution is 2.0 ml BAC water for 10mg vial. A typical research study window uses 5 - 10 mg / daily (10-20 day cycle).
For a 10mg epithalon research vial, add 2.0 ml BAC water (5.0 mg/ml). Due to the short recommended in-solution stability window (14 days), plan research cycles so that each reconstituted vial is consumed within this period. For a 10-day protocol at 10 mg/day, reconstitute one 10mg vial per day using sterile water for injection and use immediately.
